The NAD+ reading shelf
The human trials beside the animal work, with the reviews that make the argument harder.
Vinten’s review narrows the human claim
For the front of the shelf, I’d choose Vinten and colleagues, 2025. Its subject is human NAD+ precursor supplementation, and it is particularly useful on the distance between blood changes and clinical efficacy. For the underlying cellular machinery, Covarrubias and colleagues, 2021 and Ying’s review of NAD-related cellular functions supply the larger biochemical setting.
The human findings
Conze: NR and whole-blood NAD+ is the dose-response paper. Yi: NMN, blood NAD+ and walking distance asks a broader functional question. Yoshino: NMN and muscle insulin sensitivity belongs beside them, with its prediabetic, postmenopausal population kept in mind.
For the cardiovascular case, read Wang: NR tolerability in heart failure alongside Freeberg: systolic pressure and arterial stiffness. They do not measure the same outcome.
The experiments that unsettle the simple story
Camacho-Pereira: CD38 and aging mice gives the decline a mechanism. Zhang: NMN and pressure-overload heart failure in mice contains the striking survival result. Williams: vitamin B3 and glaucoma-prone mice concerns eyes rather than a general aging verdict.
Audrito: NAD+ synthesis as a melanoma vulnerability and Garrido and Djouder: NAD+ deficits, aging and cancer are the pair that prevents replenishment becoming a universal answer. Yoshino’s diabetes experiments in mice and Li’s SIRT3 review complete the animal and mechanistic portion of this list.
